YES! I was literally just looking for this when trying to tag a sci-fi I'm reading right now. I vote for both:
* Location > Setting  > Space
* Location > Setting  > Another Planet

Good idea. Maybe under Location it would be good to add another main group:

Location > Off-world

Then include specific tags: 

alien planet
deep space
human colony = permanent settlement off Earth (Mars, Moon, or other planets)
spaceship
space station
